ABSTRACT

Objective:Astragalus polysaccharide (APS) was used in combination with ionizing radiation (IR) to investigate the mechanism of APS on the radiosensitivity of human nasopharyngeal narcinoma CNE-1 cells and the epithelial-mesenchymal transition (EMT). Method:Cell counting kit-8 (CCK-8) was used to detect the cytotoxicity of different concentrations of APS (0,6.25,12.5,25,50,100,200 g·L-1) on CNE-1 cells. Colony formation assay was used to calculate the survival fraction (survival fraction, SF) of CNE-1 cells treated with 12.5 g·L-1 APS combined with different radiation doses (0,2,4,6 Gy). The linear quadratic equation mathematical model (LQ) was used to draw the radiosensitivity curve according to SF value. Cell scratch and transwell chamber test were used to detect the migration and invasion ability of cells in each group. The apoptosis of cells in each group was detected by flow cytometry, Western blot was used to detect the expressions of EMT markers, apoptosis markers and protein kinase B/extracellular regulated protein kinases (Akt/ERK) pathway proteins in each group. Result:The results of colony formation assay and radiosensitivity curve showed that the combination of non-toxic dose of 12.5 g·L-1 APS and radiation dose of 4 Gy could significantly increase the radiosensitivity of CNE-1 cells. Compared with blank group and IR group, APS combined with IR could significantly inhibit the migration and invasion of CNE-1 cells (P<0.05), and increase the rate of apoptosis (P<0.05). In addition, compared with the blank group and the IR group, APS combined with IR could significantly down-regulate the expressions of N-cadherin, p-Akt and p-ERK, and significantly up-regulate the expressions of E-cadherin, Bax and Caspase-3 (P<0.05). Conclusion:APS combined with IR can inhibit the migration and invasion of CNE-1 cells, and increase the apoptosis induced by radiotherapy, which may be related to the inhibition of EMT and Akt/ERK pathway.

ABSTRACT

<p><b>OBJECTIVE</b>To determine whether serum homocysteine and folate levels are correlated with the occurrence of neonatal asphyxia and to study the effects of gender and gestational age on serum homocysteine and folate levels.</p><p><b>METHODS</b>Thirty-five neonates with mild asphyxia (19 males and 16 females) and 40 normal neonates (control group,18 males and 22 females) were enrolled in this study. The asphyxia and the control groups consisted of 10 and 11 cases of preterm infants respectively. Serum homocysteine levels were measured using ELASA. Serum folate levels were measured using radioimmunity assay.</p><p><b>RESULTS</b>Serum homocysteine level (14.66+/-2.61 micromol/L vs 7.55+/-0.50 mumol/L; P<0.05) was significantly higher and serum folate level (2.47+/-0.24 ng/mL vs 3.28+/-0.28 ng/mL; P<0.05) was significantly lower in the asphyxia group than that in the control group. There were no significant differences in serum levels of homocysteine and folate between males and females either in the asphyxia group or the control group. The asphyxiated neonates born at premature showed increased serum homocysteine level compared with the full-term neonates with asphyxia (21.25+/-5.01 micromol/L vs 12.34+/-2. 01 micromol/L; P<0.05).</p><p><b>CONCLUSIONS</b>The increased serum homocysteine level and decreased serum folate level are correlated with the occurrence of neonatal asphyxia. Serum homocysteine and folate levels are not associated with the gender. A more significantly increased serum homocysteine level may be found in asphyxiated neonates born at premature.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the chemical constituents from the stem bark of Trewia nudiflora.</p><p><b>METHOD</b>The chemical constituents were isolated by silica gel and sephadex LH - 20 column chromatography, and the structures were elucidated by means of spectral analysis.</p><p><b>RESULT</b>Ten compounds were obtained from EtOAc fraction of EtOH extract and identified as stigmast-4-en-6beta-ol-3-one (1), stigmast-4-en-6alpha-ol-3-one (2), 7beta-hydroxysitosterol (3), 7alpha-hydroxysitosterol (4), schleicheol 2 (5), taraxerone (6), abbeokutone (7), beta-hydroxypropiovanillone (8), o-vanillyl alcohol (9), glycerol monopalmitate (10).</p><p><b>CONCLUSION</b>Compounds 1-5 and 7-9 were isolated from this plant for the first time.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of Jingui Shenqi Pill (JSP) on the expression of glucocorticoid receptor (GR) in the skin lesion and its clinical effect in treating bullous pemphigoid (BP) patients.</p><p><b>METHODS</b>Thirty BP patients were randomly divided into the treatment group (n=15) treated with JSP plus prednisone and the prednisone group (n=15) with prednisone alone both for 4 weeks. And a normal control group was set up also. Expressions of GR-alpha and GR-beta in the skin lesion of BP patients and the normal skin of the normal control were detected by immunohistochemical assay. Results The total effective rate was 93.33% in the treatment group, significantly higher than that in the prednisone group which was 73.33% (P < 0.05); GR-alpha expression was higher in the treatment group than that in other two groups (P < 0.01), while GR-beta expression in the treatment group was lower than that in the prednisone group (P < 0.01).</p><p><b>CONCLUSION</b>JSP could increase GR-alpha expression and decrease GR-beta expression in the skin lesion of BP patients, so as to improve sensitivity of skin to glucocorticoid.</p>

ABSTRACT

Objective To analyze the relation between the CT changes of ventricular volume and prognosis in newborn of perinatal hypoxic-ischemic brain damage.Methods From Jan. 2002 to Dec. 2006, 105 newborns with perinatal hypoxic-ischemic brain damage (moderate to severe) were divided into 2 groups (group A with nervous system sequel and group B without nervous system sequal). Brain CT was performed in 24 hours after admission in all cases and at 1 and 3-month time point in some cases according to their convalescence condition. Huckman index,the third ventricular width on CT images of various time points were measured and calculated by radiologist. ABSTRACT

<p><b>OBJECTIVE</b>To investigate the risk factors for postoperative pulmonary complications (PPC) after gastroduodenal operation.</p><p><b>METHODS</b>From December 1999 to December 2003, clinical data of 508 patients undergoing gastroduodenal operation were analyzed retrospectively. Risk factors for PPC were screened.</p><p><b>RESULTS</b>The complication rate of PPC was 25.8% (131/508). Multivariate logistic regression analysis revealed that age (OR=1.052), history of respiratory diseases (OR=2.915), serum albumin level (OR=0.995), length of intratracheal intubation (OR=1.005), length of nasogastric intubation (OR=1.059) and length of postoperative mechanical ventilation (OR=1.367) were risk factors for PPC.</p><p><b>CONCLUSION</b>Patients with old age, lower serum albumin level, intraoperative or postoperative nasogastric intubation, intratracheal intubation or long-term mechanical ventilation were more prone to develop PPC.</p>

ABSTRACT

Objective To study the effect of minimal enteral feeding and mechanic ventilation on motilin(MTL) in premature infants and find a new way to promote gastrointestinal motility especially in critical illness,so as to develop their nutritional condition.Methods From Nov.2004 to Jun.2006, 60 premature infants were recruited at NICU of Xi′an children′s hospital,who were assigned to minimal enteral feeding and normal feeding groups,mechanic ventilationor non-mechanic ventilation was conducted based on premature infants′ illness.Their peripheral vein blood was collected at different time respectively: before their first breast-feeding,3 and 7 days after their birth and their MTL levels was measured by radioimmunoassay.The effect of 2 factors on hormones were factorially analyzed.The difference of gastrointestinal motility between 2 feeding groups.Results Mechanic ventilation could not alter MTL levels.But minimal feeding could significantly promote motilin′s secretion(P0.05).None of patients suffered from necrotizing enterocolitis.Conclusion Minimal enteral feeding can significantly increase gastrointestinal motility of premature infants,as well as those infants who are conducted mechanic ventilation.
